Oh my.... there was no band. So the antibody binds to the beads, but the antigen is not binding to the antibody. Why why why.... 
So far...
Salmon was an IgE clone... that is why it did not bind to the protein A/G. Fortunately protein L binds to Salmon, so I can pull down the antigen.

Oh God! One simple misunderstanding, that was all I needed to put my 1 week woth of hard work down the drain. Literally, my mAb went down the drain.

I have been using Google Calender for scheduling my experiments. Recently, I felt the need for project management software, due to the complexity. I explored the internet and found a solution.
Gnome Planner.
It is simple, free, open-source, and cross-platform (works well on Windows and Linux).
I saved the data file in Dropbox and now I can access my project schedule from any computer at work or home!
I can do more work by planning well.
To Download the program, first you need GTK+ runtime for windows. This is, after all, Linux native program. Download and install GIMP for windows that comes with GTK+ runtime. Then you can install "planner-0.14.4.exe" from Gnome live site.

Deglycosylationはうまくいったのかよくわからんが、
バンドが全部消えたのでたぶん抗体はGlycoepitopeを認識するのだろう。
Isotypingもやったのだが、ELISAじゃなくてStripだったので１０分で結果が。
WhaleはMでSalmonがG2bということでSalmonで抗体つくるか。

曲がり角のレストラン
テリータウンのメイン通り
あの曲がり角にあるグリーク（ギリシャ料理）レストラン。
小さなお店だけど
酸っぱさがやみつきになるチキンレモンスープと
ステンレスのボウルに新鮮な野菜たちがどかどか山盛りのグリークサラダ、
そして愛想のいいヒゲのおじさんのウェイター。
最近の不況で小さなレストランたちがつぶれていくのだが
ここだけは生き残って欲しいとおもう。

というわけでえいかをラボへ連れて行くことに。
えいかはラボが好き。
このまえ来た時は
BALB/Cのネズミさんたちのルビーのような赤い目とか
Hela細胞を顕微鏡で見たら
「It's so beautiful. They are like diamonds.」
とか言ってたし。
今日はBumiの娘Raniaも来ていて
二人で手をつなぎながらはしゃいでいた。
Hybridomaの細胞たちを見せたら
「They are different from what I saw last time. These are like bubbles.」
とな。
ちゃんと見ている。
細胞もいろんな種類に分化したのがあるのだと説明。
えいかは細胞でできているのだというと不思議そうな顔をしていた。
そう、世の中不思議なことはいっぱいあるのだ。
そういう不思議をいっぱい感じられるような子に育ってほしい。

ケイタイ紛失してから
昔のケイタイを復活させて使っていたのだが、
賢寿のボタン連打攻撃に耐えきれず故障した様子。
画面が真っ青なのだが、普通に通話できる。
が、電話帳機能が使えないので不便だ。
2年契約が切れるのが半年以上先なので
気軽に買い替えともいかない。
うーん、eBayで安いの拾うか？
昔のケイタイを復活させて使っていたのだが、
賢寿のボタン連打攻撃に耐えきれず故障した様子。
